The most modern and highly specific RNAi tools are based on artificial sRNAs (art-sRNAs), which are computationally designed to be highly specific and bind only to the target mRNAs without interacting with other cellular RNAs. In our laboratory, we have successfully significantly reduced the size of the MIR390a precursor from Arabidopsis thaliana (AtMIR390a). Crude extracts have been obtained from plants infected with the viral vector containing the chimeric minimal precursor, and it has been possible to infect new plants and express the amiRNAs of interest by topically applying these extracts through mechanical inoculation and spraying.
