The invention relates to a method for enhancing osteogenic differentiation of MSCs by culturing them in osteogenic medium with the addition of an EphB3 receptor blocking reagent. Said blocking reagent can be selected from polyclonal antibodies, monoclonal antibodies, EphB3 blocking peptides, fusion proteins, RNAi and/or shRNA blocking EphB3 gene expression, editing of the gene encoding EphB3 by the CRISPR/CAS technique or combinations thereof. The invention also concerns both the osteogenic lineage cells obtained by the described method and the EphB3 receptor blocking reagents, for use in the treatment of bone pathologies with bone tissue deficiency and/or bone fractures. Also included are pharmaceutical compositions containing the osteogenic lineage cells obtained by the described method and/or the EphB3 receptor blocking reagents.

The invention represents a new contribution to the osteogenic differentiation of mammalian mesenchymal stromal cells procedures , which significantly improves the capacity to obtain a large amount of bone, with application in the treatment of bone diseases and/or fractures. Very good results have been obtained in the treatment of osteoporosis in animal models.
