The invention describes a device (1) for characterising the roughness profile of a tissue sample, which comprises: a laser source (2) that illuminates the tissue surface (100); a photodetector (3) that receives light backscattered by the tissue surface (100); and a movement means (4), designed to alternate between a first position in which a rotary frosted glass plate (5) is disposed in the laser beam path towards the surface (100), a second position in which a rotary medium-wave sheet (6) is disposed in the laser beam path towards the surface (100), and a third position in which neither the frosted glass plate (5) nor the medium-wave sheet (6) is disposed in the laser beam path toward the surface (100), or the medium-wave sheet (6) is disposed in a non-rotary fixed position.
