Procedure for the detection, quantification and/or identification of DNA of the Galician wheat varieties "Callobre" and "Caaveiro"

The Pan Gallego IGP prescribes the use of at least 25% Galician wheat in its preparation. Currently, two varieties of Galician wheat are registered: "Caaveiro" and "Callobre". Microsatellite markers (SSR) allow the identification and quantification of Galician wheat in biological samples, being a feasible tool in the traceability of the use of Galician wheat in the production of Galician bread covered by the IGP.
The present invention relates to the detection, quantification and/or identification of specific DNA of the 'Caaveiro' and 'Callobre' varieties from different types of samples such as fresh plant material, monovarietal flour samples, flour mixture samples, wheat grain samples, bread samples, sourdough samples or bread dough samples before baking. The invention is based on the amplification, detection and quantification of DNA using either PCR and a sequencer or real-time PCR. The proposed procedure comprises the amplification of five microsatellites located in 5 non-coding zones of the genome. In said amplification, fluorochromes are included that allow the detection of the amplification products either by fluorescence reading during the use of real-time PCR or after a PCR by means of a sequencer that allows microsatellites to be identified based on to the specific fluorescence peak. The high specificity of the primers makes it possible to identify the specific varieties.
