COLORIMERIC METHOD FOR DETECTION OF AMPLIFIED GENETIC MATERIAL AND USES THEREOF

The present disclosure relates to a colorimetric method for a detection of an amplified genetic material in a biological sample, comprising the following steps: obtaining a biological sample; amplifying the genetic material; incubating the solution of the previous step; adding murexide and a zinc(II) compound to the solution; and evaluating the color change of the solution. An aspect of the present disclosure relates to the use of murexide and a zinc(II) compound as a detection ensemble for detection of an amplified genetic material in a biological sample. A kit, a microfluidic diagnostic cartridge or a lab-on-chip device for detection of an amplified genetic material in a biological sample by the described method is also disclosed.
Recently, it was shown that SARS-CoV-2 could be detected in the saliva of infected individuals, highlighting salivary tests as valuable alternatives for COVID-19 diagnosis, Saliva-based testing has numerous advantages over NP sampling, especially in a mass screening scenario. It can be performed easily and non-invasively, thus minimizing patient discomfort, and it does not require specialized personnel or the use of protective equipment, which saves time and reduces costs. For these reasons, saliva RT-PCR and RT-LAMP tests for SARS-CoV-2 detection have been widely explored in recent months.
